Target Information
The CXCR3 gene encodes a G protein-coupled receptor selective for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant). These chemokines belong to the CXC subfamily, characterized by a single amino acid residue separating the first two conserved cysteines. Chemokine binding induces leukocyte trafficking responses such as integrin activation, cytoskeletal rearrangement, and chemotaxis. CXCR3 signaling involves Gi-class G proteins and may utilize similar pathways to other chemokine receptors. Expression is prominent in effector/memory T cells and inflamed tissues, suggesting a role in inflammatory cell recruitment. CXCR3 is a potential target for small-molecule antagonists in inflammatory disease therapy. Multiple transcript variants encoding different isoforms have been identified.
