Target Information

The CXCR3 gene encodes a G protein–coupled receptor selective for three chemokines: IP10 (interferon-γ–inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant). These chemokines belong to the CXC subfamily, characterized by a single amino acid separating the first two conserved cysteine residues.

Binding of these chemokines to CXCR3 triggers cellular responses such as leukocyte trafficking, integrin activation, cytoskeletal reorganization, and chemotaxis. The receptor couples to Gi-class heterotrimeric G proteins, and its signaling cascade resembles other chemokine receptors. Following ligand-induced internalization, receptor recycling restores responsiveness.

CXCR3 is prominently expressed in effector/memory T cells and T cells in inflamed tissues. IP10, Mig, and I-TAC are locally produced in inflammatory lesions, implicating CXCR3 in inflammatory cell recruitment. Consequently, CXCR3 is a potential therapeutic target for inflammatory disease modulation. Multiple transcript variants encoding distinct isoforms have been identified.
