Target Information

ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays an essential role in endoplasmic reticulum-based stress signaling.

IRE1 acts as a sensor of unfolded proteins in the ER, initiating the unfolded protein response by splicing XBP1 mRNA into an active transcriptional activator. ERN1 controls IRE1 proteolysis in mammalian cells and has critical functions in extraembryonic cells essential for fetal viability. Null mutants are viable but IRE1 is essential under stress conditions that cause unfolded proteins to accumulate in the ER. Depletion of IRE1p through dominant negative expression leads to apoptosis under such stress conditions.
