Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 up-regulates stress response genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is important for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays roles in:
- Preventing proliferation of damaged cells
- Acting as an anticancer agent
- Regulating responses to aging and oxidative stress
Mutations in TP53 are common in human cancers and can result in loss of tumor suppressor activity. Germline mutations are also linked to cancer-prone syndromes such as Li-Fraumeni syndrome.
