Product Specific Information
The IT2.2 monoclonal antibody reacts with human CD86, an ~80 kDa surface receptor also known as B7-2. CD86 and CD80 are members of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated through various stimuli including BCR complex, CD40, and cytokine receptors.
CD86 interacts with CD28 and CD152 (CTLA-4) on T cells, playing a critical role in T-B cell crosstalk, T cell costimulation, autoantibody production, and Th2-mediated Ig production.
The IT2.2 antibody is validated for flow cytometric analysis of normal human peripheral blood cells and can be used at ≤0.5 µg per test (100 µL final volume). Optimal titration is recommended for best results.
Target Information
CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation during antigen presentation. Insufficient co-stimulation via CD86/CD80 can lead to immune tolerance.
CD86 is a type I membrane protein in the immunoglobulin superfamily, expressed by antigen-presenting cells. Binding of CD86 with CD28 provides a stimulatory signal, while interaction with CTLA-4 inhibits T-cell activation.
Alternative splicing results in multiple isoforms. CD86 dysfunction has been associated with gallbladder squamous cell carcinoma and myocarditis.
