Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM that associates with CD18 to form Mac-1 (αMβ2 integrin). It binds CD54 (ICAM-1), C3bi, and fibrinogen, and is expressed on macrophages, NK cells, granulocytes, activated lymphocytes, and mouse B-1 cells.
This antibody cross-reacts with human CD11b and can detect the antigen on human monocytes, granulocytes, and NK cell subsets.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse bone marrow cells at ≤0.125 µg/test (100 µL final volume). Optimize titration for best performance.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended detection filter: 660/20 bandpass.

Usage Notes:

  • Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
  • Protect vial and samples from light (photo-induced oxidation sensitivity).
  • Samples can be fixed using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) and stored up to 3 days at 4°C in the dark.
  • Excitation: 405 nm; Emission: 645 nm; Laser: Violet.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that forms a heterodimer with CD18 (integrin beta-2), known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia.
CD11b/CD18 serves as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating key cell-cell and cell-matrix interactions.
Expression varies by tissue and cell type and is commonly used as a microglial marker. Dysregulation is linked to systemic lupus erythematosus and ITGAM-related susceptibility.
