Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (FcγRIII), which exists as two isoforms: FcγRIIIA and FcγRIIIB.

  • Expression: FcγRIIIA on monocytes, macrophages, NK cells; FcγRIIIB on neutrophils.
  • Function: Mediates ADCC, phagocytosis, cytokine release, and antigen presentation.
  • Epitope: 3G8 recognizes the same epitope as CB16 but distinct from B73.1.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry using human peripheral blood cells.
Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorophore Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm.
Use a 660/20 bandpass filter. Ensure the instrument detects this fluorochrome.

Staining Recommendations:
When using multiple Super Bright conjugates, employ Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions.
Protect vial and samples from light due to photo-induced oxidation sensitivity.

Fixation:
Samples may be fixed using IC Fixation Buffer (Product # 00-8222) or 1-Step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

CD16 encodes a receptor recognizing the Fc portion of IgG, facilitating immune complex clearance and cellular cytotoxicity.

  • Clinical relevance: Mutations linked to immunodeficiency 20, herpes zoster, lupus, and neonatal neutropenia.
