Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG (50–80 kDa).
CD16 includes two genes:

  • CD16A (FcγRIIIA): Found in macrophages and NK cells (transmembrane form)
  • CD16B (FcγRIIIB): Found in neutrophils (GPI-anchored form)

Polymorphisms NA-1 and NA-2 exist for CD16B, with NA-2 homozygotes showing lower phagocytic capacity.
CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, and mediates ADCC, phagocytosis, degranulation, and proliferation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for human peripheral blood cells at 5 µL (0.125 µg)/test (in 100 µL total volume, 10⁵–10⁸ cells/test). Optimal titration is recommended.

Fluorochrome Information:

  • Super Bright 645 is a tandem dye excited by 405 nm laser, emits at 645 nm
  • Recommended filter: 660/20 bandpass
  • Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes
  • Light sensitive – protect from light
  • Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark

Target Information

CD16 (FcγRIII) encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance, antibody-dependent cytotoxicity, and viral infection enhancement.

  • Gene: FCGR3A (chromosome 1), similar to FCGR3B
  • Expression:
    • FCGR3A: NK cells (transmembrane form)
    • FCGR3B: Neutrophils (PI-anchored form)
  • Mutations linked to Immunodeficiency 20, Herpes Zoster, Systemic Lupus Erythematosus, and Alloimmune Neonatal Neutropenia
  • Alternative splicing produces multiple isoforms.
