Product Specific Information

Description:
The 2H7 monoclonal antibody reacts with human CD20, a 33–36 kDa transmembrane protein expressed by developing and mature B cells, but not plasma cells. CD20 may play a role in B-cell activation and is also detected at low levels on a subset of mature T cells.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-diluted and validated by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume). Cell number can range from 10⁵ to 10⁸ cells/test.

Fluorophore Information:
Super Bright 645 is a tandem dye excited by a 405 nm violet laser and emits at 645 nm. Use a 660/20 bandpass filter. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal brightness loss. Clone-specific performance should be empirically determined.

405 nm / 645 nm (Violet Laser)

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 remains on the cell surface after antibody treatment, making it a useful target for therapeutic B-cell depletion.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
