Product Specific Information

The GL3 monoclonal antibody reacts with the gamma delta T-cell receptor (TCR) complex on all γδ TCR-bearing T cells, but not with αβ TCR. The γδ TCR is present in the thymus, epidermis, intestinal epithelium, peritoneal cavity, and lymphoid tissues.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse lymph node cells. Recommended ≤0.5 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

Pre-staining Recommendation:
Pre-stain cells with anti-mouse CD16/32 (Product #14-0161-85) to prevent non-specific Fc-mediated binding.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

TCR gamma/delta (T-cell receptor γδ) are specialized T cells involved in immune defense. TCRs distinguish foreign from self-peptides presented by MHC class II molecules, essential for adaptive immunity.
Self-reactivity of TCRs is linked to autoimmune diseases, while mutant self-peptides are associated with tumors.
Gamma-delta T cells, primarily found in epithelial tissues, exhibit limited TCR diversity and distinct antigen recognition from αβ T cells. Certain γδ T cell subsets show antitumor and immunoregulatory activity.
