Target Information

Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. ER acts as a ligand-activated transcription factor that, upon binding estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction allows ER to regulate gene transcription positively or negatively through coactivator or corepressor recruitment.
There are two forms of ER, alpha and beta, encoded by ESR1 and ESR2 respectively. At least four ER-alpha isoforms are known (66 kDa, 53 kDa, 47 kDa, 35 kDa). ERs are expressed in various tissues and are essential for sexual development, reproductive function, and bone metabolism. They are implicated in pathological processes including breast cancer, endometrial cancer, and osteoporosis.
