Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor functions as a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding regulates gene transcription through coactivator or corepressor recruitment.
There are two forms of estrogen receptors, alpha (ESR1) and beta (ESR2), encoded by separate genes. Multiple ER-alpha isoforms exist due to alternative splicing (66 kDa, 53 kDa, 47 kDa, 35 kDa).
ERs are widely expressed in various tissues, crucial for sexual development, reproductive function, and bone physiology, and are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
