Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding estrogen, undergoes a conformational change enabling dimerization and DNA binding to estrogen response elements (ERE). ER regulates gene transcription positively or negatively through coactivator or corepressor recruitment.

There are two main forms of the estrogen receptor—alpha and beta—encoded by separate genes (ESR1 and ESR2). Alternative RNA splicing produces at least four alpha isoforms (66, 53, 47, and 35 kDa). Estrogen receptors are expressed in various tissues and play critical roles in sexual development, reproductive function, and bone physiology. They are also implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
