Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding regulates gene transcription through coactivator or corepressor interactions.

There are two forms of estrogen receptors, alpha and beta, encoded by separate genes (ESR1 and ESR2). Alternative RNA splicing produces at least four ER-alpha isoforms (66 kDa, 53 kDa, 47 kDa, and 35 kDa). Estrogen receptors are widely expressed and are essential for sexual development, reproductive function, and bone maintenance. They are implicated in pathological processes including breast cancer, endometrial cancer, and osteoporosis.
