Product Specific Information

  • For Immunohistochemistry (PFA fixed):
    Incubate antibody for 30 minutes at room temperature.
    Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and the nucleolus. Ki-67 shows cell cycle–dependent localization: perinucleolar expression at G1, nuclear matrix expression at G2, and chromosomal expression during M phase.

Ki-67 is widely used as a proliferation marker because it is absent in G0 cells and increases from G1 through mitosis. Ki-67 antibodies help determine the proliferative fraction of cells in neoplasms, correlating with tumor grade and prognosis. It is also used as a neuronal marker for cell cycling and proliferation.
