• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly during late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Ki-67 exhibits cell cycle-dependent localization: perinucleolar expression at G1, nuclear matrix expression at G2, and chromosomal association during M phase.
As Ki-67 is absent in G0 cells and increases from G1 through mitosis, it serves as a reliable proliferation marker. Ki-67 antibodies help determine the cell growth fraction in neoplasms, showing strong correlation between low Ki-67 index and histologically low-grade tumors. It is routinely used as a neuronal marker for cell cycling and proliferation.
