• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that enables interaction with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Ki-67 shows cell cycle-dependent localization: perinucleolar in G1, nuclear matrix in G2, and chromosomal during M phase.
Because Ki-67 is absent in G0 cells and increases throughout the cell cycle, it serves as a reliable proliferation marker. Ki-67 antibodies are used to determine the cell growth fraction in neoplasms, with prognostic value comparable to the thymidine-labeling index. Low Ki-67 index correlates with histologically low-grade tumors.
It is routinely used as a neuronal marker for cell cycling and proliferation.
