Antibody detects endogenous levels of TSC1 only when phosphorylated at Ser505.

Target Information

Tuberous sclerosis complex (TSC) is an autosomal dominant tumor syndrome caused by mutations in either of the TSC1 or TSC2 tumor suppressor genes.
The products of these genes form a protein complex that indirectly decreases signaling of the mammalian Target of Rapamycin (TOR), a conserved serine/threonine kinase regulating cell growth and cycle through nutrient and growth factor signals.
TOR activity is stimulated by Rheb, a Ras superfamily G-protein, when the GTP/GDP ratio bound to Rheb is high.
Immunoprecipitated TSC1/TSC2 stimulates Rheb GTPase activity in vitro, decreasing Rheb-mediated TOR activation.
Overexpression of TSC1 and TSC2 reduces the GTP/GDP ratio bound to Rheb and inhibits cell growth.
A shorter 40 kDa isoform of TSC1 exists, but its function remains unknown.
