Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of blood plasma proteins. It inhibits amyloid fibril formation by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants in vitro.
Clusterin does not require ATP and maintains partially unfolded proteins suitable for refolding by other chaperones such as HSPA8/HSC70. It does not refold proteins by itself.
Binding to cell surface receptors triggers internalization and subsequent lysosomal or proteasomal degradation. Secreted isoform 1 protects cells against apoptosis and complement-mediated cytolysis.
Intracellular isoforms interact with ubiquitin and SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complexes, promoting ubiquitination and proteasomal degradation of target proteins.
Clusterin modulates NF-kappa-B transcriptional activity, with nuclear isoforms promoting apoptosis and mitochondrial isoforms inhibiting BAX-dependent cytochrome c release, thereby suppressing apoptosis.
It plays an important role in cell proliferation regulation.
