Product Specific Information
This LUVNKLA monoclonal antibody recognizes human and mouse STAT3 when phosphorylated on Tyr705.
STAT family proteins (STAT1, 2, 3, 4, 5A, 5B, 6) regulate apoptosis, differentiation, and proliferation in a cytokine-specific manner.
STAT3 is activated downstream of cytokines such as IFNs, IL-5, IL-6, IL-10, and LIF, and plays a key role in Th17 differentiation, cell growth, and survival.
Loss-of-function mutations reduce IL-6/IL-2-mediated proliferation, while hyperactivation is associated with poor cancer prognosis.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested for intracellular staining of stimulated human peripheral blood cells.
Use 5 µL (0.125 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).
Recommended Protocol:
- Use Protocol C: Two-step Fixation/Methanol.
- Protocol A (cytoplasmic) and Protocol B (nuclear) are not applicable.

STAT3 is a member of the STAT protein family activated by receptor-associated kinases in response to cytokines and growth factors.
Phosphorylation at Tyr705 by JAK1/JAK2 is essential for dimerization, nuclear translocation, and DNA binding.
STAT3 mediates transcription of genes involved in cell growth and apoptosis and interacts with cofactors such as CREB-binding protein and c-Jun.
The STAT3 gene is located on chromosome 17q, and knockout models show embryonic lethality, highlighting its essential role in development.
