Product Specific Information

Description:
The PO3.1 monoclonal antibody reacts with mouse CD86, an ~80 kDa surface receptor also known as B7-2. CD86 and CD80 are members of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated through stimuli such as BCR complex, CD40, and cytokine receptors. It is also expressed by activated mouse T cells and thioglycolate-elicited peritoneal cells.

CD86 interacts with CD28 and CD152 (CTLA-4) on T cells, playing a critical role in T-B cell communication, costimulation, autoantibody production, and Th2-mediated Ig production. The kinetics of CD86 upregulation suggest its major role during the primary phase of immune response.

Applications Tested:
The PO3.1 antibody has been tested by flow cytometric analysis of resting and activated mouse splenocytes and can be used at 0.25 µg per test (defined as the amount that stains a sample in 100 µL). Cell number may vary from 10⁵ to 10⁸ cells/test. Optimal performance should be determined empirically.

Target Information

CD86 is one of two ligands (along with CD80) for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation during antigen presentation. Insufficient co-stimulation involving CD86/CD80 can induce tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed by antigen-presenting cells, and serves as a ligand for CD28 and CTLA-4 on T cells.
Binding of CD86 with CD28 provides a costimulatory signal for T-cell activation, while binding with CTLA-4 negatively regulates activation and reduces immune response.
Alternative splicing results in two transcript variants encoding different isoforms of CD86. Diseases associated with CD86 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
