Product Specific Information

The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor belonging to the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells and is upregulated upon stimulation through BCR, CD40, and cytokine receptors.
CD86 interacts with CD28 and CTLA-4 on T cells, playing a crucial role in T-B cell crosstalk, T cell costimulation, and antibody production. Its rapid upregulation supports its role in the primary immune response.

Applications Reported: Flow cytometry and functional blocking studies.
Applications Tested: Flow cytometric analysis of human peripheral blood cells. Recommended ≤0.25 µg/test (100 µL volume). Optimize antibody titration for each assay.

Target Information

CD86 is one of two ligands (with CD80) for CTLA-4 and CD28, functioning as a costimulatory molecule during antigen presentation. Insufficient CD86/CD80 co-stimulation can induce tolerance.
CD86 is a type I membrane protein and member of the immunoglobulin superfamily, expressed by antigen-presenting cells.
Binding with CD28 activates T cells, while interaction with CTLA-4 inhibits activation.
Alternative splicing yields multiple isoforms.
Associated diseases include gallbladder squamous cell carcinoma and myocarditis.
