Product Specific Information

Antibody detects endogenous levels of p53 only when phosphorylated at Serine 15.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, regulating G1 or G2/M arrest.
The protein consists of an N-terminal transactivation domain, a DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain.
Phosphorylation at Ser15 is crucial for p53 activation and stabilization. p53 functions as an anticancer agent by blocking proliferation of damaged cells. Phosphorylation at Ser392 is associated with tumor formation.
Additionally, p53 is linked to aging, oxidative stress, and cognitive functions such as learning and memory. Normally expressed at low levels, p53 is highly expressed in transformed cell lines, contributing to malignancy. Mutations in TP53 result in loss of tumor suppressor activity and occur in many cancers, including hereditary cancer syndromes such as Li-Fraumeni syndrome.
