Target Information
Huntington’s disease (HD) is a neurodegenerative disorder characterized by loss of striatal neurons, caused by expansion of a polyglutamine tract in the huntingtin protein. HAP1 was identified through two-hybrid library screening; its binding to huntingtin correlates with polyglutamine tract expansion. HAP1 interacts with cytoskeletal proteins such as dynactin and pericentriolar autoantigen protein 1, suggesting a role in vesicular trafficking or organelle transport. It also participates in huntingtin-enhanced BDNF transport along cellular microtubules. Impaired transport leads to reduced neurotrophic support and neuronal toxicity, potentially contributing to pathogenesis. Several alternatively spliced isoforms of HAP1 have been described.
