Target Information
Isocitrate dehydrogenases catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes are divided into NAD(+)-dependent and NADP(+)-dependent subclasses.
Five isocitrate dehydrogenases have been reported:
- Three NAD(+)-dependent forms localized to the mitochondrial matrix
- Two NADP(+)-dependent forms, one mitochondrial and one cytosolic
The IDH1 gene encodes the NADP(+)-dependent isocitrate dehydrogenase found in cytoplasm and peroxisomes. It includes a PTS-1 peroxisomal targeting sequence and contributes to NADPH regeneration for peroxisomal reduction reactions and cytoplasmic NADPH production.
