Cysteine protease ATG4B is an enzyme encoded by the ATG4B gene.
Autophagy is a key cellular process controlling the turnover of damaged organelles and proteins.
ATG4B acts as a deubiquitin-like protease, cleaving the C-terminal amino acid of ATG8 family proteins (MAP1LC3, GABARAPL1, GABARAPL2, GABARAP) to expose a C-terminal glycine.
This glycine exposure enables conjugation to phosphatidylethanolamine (PE) and membrane insertion, essential for autophagy.
ATG4B is mainly expressed in skeletal muscle, brain, heart, liver, and pancreas.
