Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 plays a key role in peripheral tolerance and autoimmune disease regulation in mice. It is mainly expressed on activated T and B lymphocytes and interacts with PD-L1 (B7-H1) and PD-L2 (B7-DC).
J43 inhibits the binding of PD-L1-Ig and PD-L2-Ig to PD-1 transfected cells and can modulate immune responses in vivo.
This antibody is validated for flow cytometric analysis of mouse splenocytes, recommended at ≤0.5 µg/test. Optimal titration is advised.
Brilliant Violet™ 605 (BV605) emits at 605 nm and is designed for cytometers equipped with a violet (405 nm) laser.
For multi-color staining, use Super Bright Complete Staining Buffer or Brilliant Stain Buffer™ to minimize non-specific polymer interactions.
- Light sensitivity: Protect from light to prevent photo-induced oxidation.
- Fixation: Samples can be stored up to 3 days at 4°C in IC Fixation Buffer or 1-step Fix/Lyse Solution. BV605 is not compatible with methanol-based fixation.

Target Information
PD-1 (CD279) is an inhibitory receptor expressed on activated T cells, mediating immune downregulation upon binding to its ligands PD-L1 and PD-L2. This interaction limits T-cell activation in the absence of danger signals. PD-1 deficiency leads to hyperactivation and autoimmune manifestations in mice. PD-1 often migrates at a higher molecular weight in SDS-PAGE despite its predicted size.
