• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Ki-67 is a nuclear protein expressed during the late G1, S, G2, and M phases of the cell cycle. It contains a forkhead-associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and the nucleolus.
Expression pattern:

• G1: Perinucleolar expression
• G2: Nuclear matrix expression
• M phase: Chromosomal expression

Ki-67 serves as a reliable proliferation marker since it is absent in G0 cells and increases progressively through mitosis. It is used to determine the growth fraction in neoplasms and has strong prognostic correlation with tumor grade. Commonly utilized as a neuronal marker for cell cycling and proliferation.
