Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly during late G1, S, G2, and M phases. The protein contains a forkhead-associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and the nucleolus.

Ki-67 shows cell cycle-dependent distribution:

  • Perinucleolar expression at G1
  • Nuclear matrix localization at G2
  • Chromosomal association during M phase

Because Ki-67 is absent in G0 cells and increases from G1 through mitosis, it serves as a reliable proliferation marker. Ki-67 antibodies are useful for determining the growth fraction of neoplastic cells, showing strong correlation between low Ki-67 index and low-grade tumors. It is routinely used as a neuronal marker for cell cycling and proliferation.
