• Immunohistochemistry (PFA-fixed): Incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.

• Ki-67 shows cell cycle-dependent distribution:
  • Perinucleolar expression at G1
  • Nuclear matrix localization at G2
  • Chromosomal association during M phase
• Not detected in G0 cells; expression increases from G1 through mitosis
• Commonly used as a proliferation marker to assess the growth fraction in neoplasms
• Low Ki-67 index correlates with low-grade tumors
• Routinely used as a neuronal marker of cell cycling and proliferation
