Target Information

RIPK1 is a serine-threonine kinase that mediates inflammatory and programmed necrosis signaling following activation by death receptors, pathogen recognition receptors (PRRs), and DNA damage. Upon TNFR1 activation by TNF-alpha family cytokines, TRADD and TRAF2 are recruited to the receptor.
RIPK1 phosphorylates DAB2IP at Ser-728, activating the MAP3K5-JNK apoptotic cascade. Ubiquitination by TRAF2 via Lys-63-link chains enhances downstream signaling through the MAPK and NF-kappa-B pathways, leading to expression of inflammatory genes.
Polyubiquitinated RIPK1 binds IKBKG/NEMO, a key regulatory subunit of the IKK complex, facilitating NF-kappa-B activation. Interaction with other RHIM-containing adapters triggers gene activation and cell death. RIPK1 and RIPK3 association forms a necrosis-inducing complex.
