Target Information

SMAD2 (MADH2, MAD2) regulates multiple cellular processes, including cell proliferation, apoptosis, and differentiation. It interacts with TGF-beta receptors through the SMAD anchor for receptor activation, enabling nuclear translocation—a central event in TGF-beta signaling. Phosphorylation of threonine 8 in the MH1 domain by ERK1 enhances transcriptional activity, negatively regulated by calmodulin. Upon TGF-beta stimulation, SMAD2 is phosphorylated, dissociates from SARA, and associates with SMAD4 for nuclear translocation and transcriptional regulation. SMAD2 can also mediate activin signaling via activin type 1 receptor kinase. Alternatively spliced transcript variants encoding SMAD2 have been observed.
