Target Information

The SMAD1 protein belongs to the SMAD family, which includes proteins similar to the Drosophila gene mothers against decapentaplegic (Mad) and the C. elegans gene Sma. SMAD proteins are signal transducers and transcriptional modulators mediating multiple signaling pathways.
SMAD1 mediates bone morphogenetic protein (BMP) signaling involved in cell growth, apoptosis, morphogenesis, development, and immune responses. Upon BMP ligand binding, SMAD1 is phosphorylated by BMP receptor kinase and forms a complex with SMAD4 to regulate transcription.
It is also targeted by SMAD-specific E3 ubiquitin ligases such as SMURF1 and SMURF2, leading to ubiquitination and proteasome-mediated degradation. Alternatively spliced transcript variants encoding the same protein have been observed.
