Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is tonsil tissue.
Target Information
ZAP-70 is a cytosolic protein tyrosine kinase belonging to the Syk family. It is expressed exclusively in T-cells and natural killer cells and plays a key role in T-cell receptor activation.
Upon TCR engagement, ZAP-70 is phosphorylated on tyrosines 292, 315, and 319 in the interdomain B region.
Phosphorylation of tyrosines 315 (Vav binding site) and 319 (Lck binding site) enhances ZAP-70 function in lymphocyte signaling, while tyrosine 292 phosphorylation attenuates activation.
