PA3-030A 항체는 human 및 mouse 조직에서 inducible nitric oxide synthase (iNOS)를 검출합니다. 이 항체는 endothelial NOS (eNOS) 또는 brain NOS (bNOS)를 검출하지 않습니다.

• Western blot 및 Immunohistochemistry (paraffin, frozen)에서 검증됨
• Western blot에서 유도된 RAW 264.7 세포 추출물 내 약 130 kDa의 iNOS 단백질 검출
• Immunogen은 mouse macrophage NOS의 C K(1131) K G S A L E E P K A T R L(1144) 서열 기반 synthetic peptide
• Blocking peptide PEP-008과 함께 사용 가능
• 이전 Thermo Scientific Cellomics High Content Screening Kit의 일부로 판매되던 항체로, 현재는 단독 연구용으로 권장됨

Target Information

iNOS (Inducible Nitric Oxide Synthase, type II)는 세포 간 신호전달에 관여하는 무기성 자유 라디칼인 NO 생성을 매개합니다.
NO는 혈관이완, 신경전달, 세포독성 등 다양한 생리적 반응에 영향을 미칩니다.

• iNOS는 간에서 발현되며, LPS 및 특정 사이토카인에 의해 유도됨
• NOS 효소군은 L-arginine을 산화시켜 L-citrulline과 NO를 생성
• 세 가지 주요 isoform 존재: nNOS (type I), eNOS (type III), iNOS (type II)
• iNOS는 대식세포, 간세포, 활막세포, 평활근세포 등 다양한 세포에서 발견됨
• IFN-γ, TNF, IL-1, IL-2, LPS 등에 의해 iNOS 발현 증가
• 단백질 키나아제 C 자극제 또한 iNOS 활성 증가 유도
• 세포 유도 후 NO 생산이 지연되어 장시간 지속적 증가
• iNOS 관련 질환: achalasia, impotence 등
