Product Specific Information
PA3-033 detects cytochrome P450 4A (isoforms 1, 2, and 3) from human, feline, rabbit, and rat tissues.
It has been successfully used in Western blot, ICC/IF, and IHC procedures.
By Western blot, this antibody detects an approximately 50 kDa protein representing P450 4A in rat liver extract.
Immunohistochemical staining of P450 4A in rat brain with PA3-033 results in perivascular and surrounding cortex staining.
The immunogen is a synthetic peptide corresponding to residues D(431)PSRFAPDSPRHS(445) of rat Cytochrome P450 4A.
Target Information
Cytochrome P450 (P450) enzymes are part of the systems that metabolize arachidonic acid to regulators of vascular tone.
P450 enzymes are monooxygenases requiring cofactors such as NADPH and P450 reductase.
Over 200 cDNAs encode P450 proteins.
Epoxygenases metabolize arachidonic acid to epoxyeicosatrienoic acids (EETs), while ω-hydroxylases produce 19- and 20-hydroxyeicosatetraenoic acids (HETEs).
20-HETE is generated by the 4A family of P450 proteins, which includes at least 8 closely related isoforms (4A1, 4A2, 4A3).
These have been cloned from liver, kidney, and testis, and detected in renal, hepatic, and brain microvessels.
