Target Information

Membrane-associated guanylate kinase (MAGUK) family members localize to the plasma membrane and act as molecular scaffolds for multi-protein complexes.
The MAGUK family includes mammalian proteins related to the Drosophila tumor suppressor discs-large (dlg) gene product, such as postsynaptic proteins, GKAPs, tight junction proteins (ZO-1–3), and CARD proteins (CARD 6, 8–12, 14).

CARD14 is a 1,004 amino acid protein with:

• N-terminal CARD domain
• Central coiled-coil domain
• C-terminal tripartite domain (PDZ, SH3, and GUK domains)

CARD14 is expressed in the placenta, where it positively regulates apoptosis and controls NF-κB activation by phosphorylating BCL10.
Epigallocatechin-3-gallate (EGCG), a polyphenol, induces CARD14 expression.
