Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
The S100A8/A9 heterocomplex, also known as MRP-8/MRP-14 complex or calprotectin, constitutes about 60% of the cytoplasmic protein fraction of circulating polymorphonuclear granulocytes and is also found in monocytes, macrophages, and ileal tissue eosinophils.
Peripheral blood monocytes carry the antigen both extra- and intracellularly, while neutrophils only intracellularly.
This complex has antibacterial, antifungal, immunomodulating, and antiproliferative effects, and acts as a potent chemotactic factor for neutrophils.
Plasma concentrations increase in diseases with elevated neutrophil activity, such as inflammatory bowel disease.
Calprotectin is detectable in feces and elevated levels have been observed in plasma, saliva, gingival crevicular fluid, stools, and synovial fluid during infection and inflammation.
