Target Information
The p90 ribosomal S6 kinases (RSKs) are a family of serine/threonine kinases at the terminus of the ERK pathway. In humans, four isoforms (RSK1–4) exist. Each RSK contains two functional kinase domains activated sequentially by phosphorylation.
RSKs regulate gene expression by phosphorylating transcriptional regulators such as c-Fos, estrogen receptor, NFκB/IκBα, and CREB. ERK activates the C-terminal kinase, which then activates the N-terminal kinase.
RSK proteins localize to both cytoplasm and nucleus. Upon growth factor stimulation, cytosolic RSK translocates to the nucleus, leading to phosphorylation and activation of transcription factors.
