Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein, which exists in two isoforms (345 and 395 kDa) expressed in dividing cells. Ki-67 is present during active cell cycle phases (G1, S, G2, and mitosis) and absent in resting cells (G0). During interphase, it localizes to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3. Ki-67 expression serves as a marker for proliferating cell fractions in tumor studies.

This antibody also recognizes canine Ki-67.

Applications Reported:
Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-diluted and validated for intracellular staining of stimulated human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended usage is 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is suitable for UV (355 nm) laser-equipped cytometers. Protect from light due to photo-oxidation sensitivity.

Buffer Recommendations:
When using multiple Brilliant Ultra Violet™ or polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (#SB-4401) or Brilliant Stain Buffer (#00-4409-75) to reduce non-specific polymer interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (#00-8222) or 1-step Fix/Lyse Solution (#00-5333) with minimal fluorescence loss. Compatible with short-term methanol fixation (<1 hour).

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin and heterochromatin regions and exhibits phase-dependent localization. As it is absent in G0 cells, Ki-67 serves as a reliable proliferation marker. In tumors, Ki-67 index correlates with growth rate and histological grade, making it a key marker in neoplastic and neuronal proliferation studies.
