Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j (ILT2, LILRB1, LIR1), a member of the ILT/LIR/MIR family.
CD85j is a single transmembrane glycoprotein with a long cytoplasmic domain containing 4 ITIMs that signal through SHP-1 interactions.
Expression is found on myeloid cells (monocytes, dendritic cells) and some lymphoid cells (NK, T, B cells).
It is correlated with leukemias (ALL, CLL) and viral infections (HIV, EBV, CMV).
Ligands include MHC Class I molecules such as HLA-G, A, F, B27, E, and F.

HP-F1 reduces CD16-dependent cytolytic activity of NK cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells.
Use at 5 µL (1.0 µg) per test (final volume 100 µL). Cell number: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm; use with UV (355 nm) laser.
Ensure instrument compatibility.

When using multiple polymer dye-conjugated antibodies, use:

  • Super Bright Complete Staining Buffer (Product # SB-4401)
  • Brilliant Stain Buffer (Product # 00-4409-75)

Light Sensitivity: Protect vial and samples from light.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).
Store fixed samples up to 3 days at 4°C in dark.
BUV737 is compatible with short-term methanol fixation (<1 hour).

Target Information

CD85j belongs to the leukocyte immunoglobulin-like receptor (LIR) family located at chromosome 19q13.4.
It encodes a transmembrane receptor with extracellular immunoglobulin domains and cytoplasmic ITIM motifs.
Expressed on immune cells, it binds MHC class I molecules and transduces inhibitory signals to regulate immune response, inflammation, and cytotoxicity.
Multiple transcript variants exist.
