Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Murine TRAIL. Anti-Murine TRAIL-specific antibody was purified by affinity chromatography employing an immobilized Murine TRAIL matrix.

Sandwich ELISA:
To detect Murine TRAIL by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen-affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Murine TRAIL (500-P303Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Murine TRAIL.

Western Blot:
To detect Murine TRAIL by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Murine TRAIL is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

TRAIL is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells but does not appear to kill normal cells, although expressed in most normal tissues.

TRAIL binds to several members of the TNF receptor superfamily, including TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and possibly TNFRSF11B/OPG.

Its activity may be modulated by decoy receptors (TRAILR3, TRAILR4, OPG) that cannot induce apoptosis. Binding of TRAIL to its receptors triggers activation of MAPK8/JNK, caspase 8, and caspase 3.

Receptors for TRAIL include two death domain-containing receptors (DR4 and DR5) and two decoy receptors (DcR1 and DcR2). TRAIL is a type II membrane protein expressed in various human tissues and induces apoptosis and NF-κB activation similar to TNF and Fas ligand.
