TRADD (Tumor necrosis factor receptor type 1-associated DEATH domain protein)은 FAS 및 TNFR1 관련 세포사멸 신호 전달의 중요한 매개체입니다. 핵 내 형태는 TRIP12에 의한 CDKN2A의 p19ARF/ARF 아이소폼의 유비퀴틴화 및 분해를 방지하여 종양 억제 인자로 작용합니다. TRADD는 TRIP12와 상호작용하여 TRIP12와 CDKN2A의 p19ARF/ARF 간 상호작용을 방해합니다. 또한 TRADD는 TNFRSF1A/TNFR1의 활성화된 세포질 도메인에 특이적으로 결합하여 FADD와의 상호작용을 매개하는 어댑터 분자로 작용합니다. TRADD의 과발현은 TNF에 의해 유도되는 두 가지 주요 반응, 즉 세포사멸(apoptosis)과 NF-kappa-B 활성화를 유도합니다.
