Description:
The HY101 monoclonal antibody reacts with glycoprotein VI (GPVI), a 62-kDa type-I transmembrane glycoprotein expressed on mature megakaryocytes and platelets. GPVI associates with the Fc receptor (FcR) gamma-chain to form a high-affinity receptor for collagen and can also bind convulxin (Cvx). Phosphorylation of the ITAMs on the FcR gamma-chain recruits Src family kinases and activates downstream signaling pathways. GPVI plays a critical role in platelet activation and aggregation.

Applications Reported:
HY101 antibody is reported for use in flow cytometric analysis and western blotting.

Applications Tested:
Tested by flow cytometric analysis and immunoblotting. For flow cytometry, use ≤0.25 µg per test (defined as the amount that stains a sample in 100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Optimal performance requires titration for each assay.

Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Glycoprotein VI (GP6) is a 58-kDa platelet membrane glycoprotein essential for collagen-induced platelet activation and aggregation. Upon vascular injury, collagen exposure triggers platelet adhesion and activation, leading to the formation of a hemostatic plug. Collagen types I, III, and VI are the primary thrombogenic components in blood vessels. This process is protective under normal conditions but can promote thrombus formation after atherosclerotic plaque rupture, contributing to myocardial infarction or stroke.
