This R3-5G4 monoclonal antibody recognizes human Runx3, a 44 kDa transcription factor belonging to the Runt domain-containing family. Runx3 is expressed in hematopoietic cells (T cells, B cells, myeloid cells) and in non-hematopoietic tissues such as epidermis, bone, and dorsal root ganglia. It plays a critical role in thymocyte development by promoting CD8⁺ T cell differentiation through Cd4 silencing. Runx3 also exhibits tumor suppressive activity in various cancers including gastric and breast cancer.

Applications Reported:
This antibody has been reported for use in western blotting and immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections.

Applications Tested:
Tested by western blotting using Raji cell line and by IHC of human skin tissue with IHC Antigen Retrieval Solution - High pH (cat 00-4956). Recommended usage ≤ 5 µg/test (100 µL final volume). Cell count range: 10^5–10^8 cells/test. Optimal performance requires titration per assay.

Target Information

This gene encodes a member of the runt domain-containing family of transcription factors. A heterodimer of this protein and a beta subunit forms a complex binding to the core DNA sequence 5'-PYGPYGGT-3', found in various enhancers and promoters, enabling transcriptional activation or suppression. It interacts with other transcription factors and functions as a tumor suppressor. The gene is frequently deleted or silenced in cancer, and multiple transcript variants encoding distinct isoforms have been identified.
