Product Specific Information
The antibody was affinity-purified from rabbit antiserum by affinity chromatography using an epitope-specific immunogen. The purity is greater than 95% as determined by SDS-PAGE.
Target Information
Vacuolar-type H⁺-ATPase (V-ATPase) is a multisubunit enzyme responsible for acidification of eukaryotic intracellular organelles.
It pumps protons against an electrochemical gradient, while F-ATPases reverse this process to synthesize ATP.
V-ATPase consists of a peripheral V1 domain (ATP hydrolysis) and an integral V0 domain (proton translocation).
The V1 domain comprises nine subunits (A–H), and the V0 domain includes five subunits (a, d, c, c′, and c″).
The V-ATPase V1 B subunit has two isoforms:
- B1 isoform: functions in proton secretion in the inner ear, maintaining endolymph pH and normal auditory function.
- B2 isoform: expressed in kidney and osteoclasts.
Mutations in the human V-ATPase B1 gene (chromosome 2cen-q13) cause distal renal tubular acidosis with sensorineural deafness.
