Product Specific Information

The DATK32 monoclonal antibody binds to a combinatorial epitope of mouse LPAM-1 (alpha4beta7), a heterodimer of the 154 kDa alpha4 subunit and the 130 kDa beta7 subunit. In mouse, the beta7 subunit is expressed on most mature lymphocytes, some thymocytes, and bone marrow cells. LPAM-1 plays a role in lymphocyte transendothelial migration across high endothelial venules (HEV), binding ligands such as MAdCAM-1, VCAM-1, and fibronectin.

Binding of DATK32 has been shown to induce aggregation of CD8+ T cell lymphoma TK1 and block LPAM-1-mediated adhesion.

Applications Reported:
Suitable for flow cytometric analysis. For functional assays, use Functional Grade purified DATK32 (Product #16-5887-85).

Applications Tested:
Validated by flow cytometric analysis of mouse splenocytes. Recommended ≤1 µg/test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test. Antibody titration is advised for optimal performance.

Target Information

Integrin beta 7 associates with integrin alpha 4 (CD49d) to form LPAM-1, expressed on intraepithelial lymphocytes. It also pairs with alpha E (CD103) to form HML-1, found on T cells near mucosal epithelium.
Ligands: VCAM-1 (CD106), MAdCAM-1, fibronectin (LPAM-1); E-cadherin (CD324, HML-1).
Integrin beta 7 mediates leukocyte adhesion and transmigration during inflammation.
