The FIB504 monoclonal antibody reacts with human and mouse integrin beta 7, a 130 kD glycoprotein that forms part of integrin complexes such as alpha4beta7 (LPAM-1) and alphaEbeta7 (HML-1).
- LPAM-1 (α4β7): Expressed on intraepithelial lymphocytes; ligands include VCAM-1 (CD106), MAdCAM-1, and fibronectin.
- HML-1 (αEβ7): Expressed on T cells near mucosal epithelium; ligand is E-cadherin (CD324).
Integrin beta 7 is essential for leukocyte adhesion and transmigration during inflammatory responses. The FIB504 antibody has reported blocking activity.
Applications Reported: Flow cytometric analysis and functional assays.
Applications Tested: Validated for flow cytometric analysis of human peripheral blood cells at ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration recommended for specific assays.
Target Information
Integrin beta 7 associates with:
- Integrin alpha 4 (CD49d) → forms α4β7 (LPAM-1)
- Integrin alpha E (CD103) → forms αEβ7 (HML-1)
These complexes mediate cell adhesion to VCAM-1, MAdCAM-1, fibronectin, and E-cadherin, facilitating leukocyte migration across endothelium during inflammation.
