Product Specific Information
Reconstitute the lyophilized powder with deionized water (or equivalent) to an antibody concentration of 0.5 mg/mL.
Akt (protein kinase B, PKB) is a serine/threonine kinase involved in key signal transduction pathways regulating cell proliferation, apoptosis, angiogenesis, and metabolism. Activation occurs via phosphorylation at two regulatory residues: threonine (by PDK1) and serine (by PDK2). Phosphorylation at Thr308 and Ser473 activates Akt α; Thr309 and Ser474 activate Akt β1/β2; and Thr305 activates Akt γ isoforms.
Akt promotes cell survival by inhibiting apoptosis through phosphorylation and inactivation of targets such as Bad, forkhead transcription factors, c-Raf, and caspase-9. Its activation is negatively regulated by PTEN and SHIP phosphatases.
This rabbit polyclonal antibody was developed using a synthetic peptide corresponding to residues surrounding Ser473 of human, rat, and mouse Akt α (PKBα, Akt1, RACα). The peptide sequence is identical to regions surrounding Ser474 of Akt β (Akt2) and Ser472 of Akt γ (Akt3).
Target Information
AKT1 (PKB alpha) is a serine/threonine kinase that regulates cell survival and proliferation. Activated AKT1 inhibits apoptosis and promotes cell cycle progression by phosphorylating multiple targets, including forkhead transcription factors and caspase-9. Activation occurs in response to insulin, PI3K, IGF1, and other growth factors. The AKT pathway is a major focus in cancer drug discovery.
