Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human Klotho. Anti-Human Klotho-specific antibody was purified by affinity chromatography employing an immobilized Human Klotho matrix.

Sandwich ELISA:
To detect Human Klotho by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. In conjunction with PeproTech Biotinylated Anti-Human Klotho (500-P296Bt) as a detection antibody, it allows detection of at least 0.2–0.4 ng/well of Recombinant Human Klotho.

Western Blot:
To detect hKlotho by Western Blot, use at 0.1–0.2 µg/mL. In conjunction with compatible secondary reagents, the detection limit for Recombinant hKlotho is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

KLOTHO is a systemic anti-aging hormone within the glycosidase1 superfamily. It encodes a type I membrane protein abundant in kidney and brain. In mice, KLOTHO deficiency leads to phenotypes resembling human aging such as arteriosclerosis, osteoporosis, and skin atrophy, along with growth retardation, short life-span, and infertility. Overexpression of KLOTHO extends life span by inhibiting insulin/IGF1 signaling. It regulates calcium/phosphorus homeostasis by inhibiting active vitamin D synthesis and is identified as a potential tumor suppressor.
